Inhibitory Effect of Ethylene Diamine Tetra Acetic Acid (EDTA), p-Chloromercuri Benzoic Acid (p-CMBA), N-Ethyl Maleimide (NEM) on Arginase Activity of the Sheep Kidney Tissue and Kinetic of Inhibition

Abstract (2. Language):
In this study, the inhibition effects of EDTA, p-CMBA, NEM on arginase activity in sheep kidney tissue were studied. Arginase activity was spectrophotometrically measured using the Tiyosemicarbazid- Diacetylmonoksim-Urea Method. Protein was determined by the method of Lowry. The kidney tissues of sheep in the present study were obtained from the Elkas sloughterhause in Elazıg. This study was carried out using 20 sample of kidney tissue. In conclusion, arginase activity in sheep kidney tissue was determined to be inhibited competative inhibition by EDTA but to be inhibited noncompetative inhibition by NEM, p-CMBA.

Bu çalışmada koyun böbrek doku arginazı üzerine EDTA, NEM, p-CMBA' in inhibisyon etkileri incelenmiştir. Arginaz aktivitesi Tiyosemikarbazid-Diasetilmonoksim-Üre metodu kullanılarak spektrofotometrik olarak ölçülmüştür. Protein Lowry metoduna göre saptanmıştır. Koyun böbrek dokuları Elazığ Elkas Kesimevi'nden temin edilmiştir. Araştırmada 20 tane koyun böbrek dokusu kullanılmıştır. Sonuç olarak, koyun böbrek doku arginazını EDTA' nın kompetetif tipte; NEM, p-CMBA' in ise nonkompetetif tipte inhibisyona uğrattığı saptanmıştır.
