Objective: This study was designed to test the effects of a long-acting somatostatin analogue SMS 201-995 (Octreotide) and omeprazole, a proton pomp inhibitor, on experimental intestinal obstruction in rat model. Methods: Albino male rats (average weight, 150-200 gr) were used for experiment. Rats were fasted for 24 hours. After they were anesthetized by inhalation with ether, the laparotomy were made for construction of a 10 cm closed-loop of terminal ileum. The rats were randomized to receive SMS 201-995 (Octreotide) 50 µg/kg (n:10), saline 0.5 ml (n:10) and omeprazole 7 mg/kg (n:10) subcutanously every 8 hours. Omeprazole was given only one dose, the other 2 doses were given as saline. Twenty four hours after the first dose of treatment the rats were sacrificed and the small bowel removed. The weight of closed-loops and the volume of luminal contents were measured. pH and electrolytes (sodium, potassium, chloride) were analysed and the loops were examined histologically. Results: No significant changes in intestinal loop weight, luminal volume, sodium, potassium, chloride density, pH level and the gross and microscopik pathologic features of intestinal distension were observed with somatostatin analogue and omeprazole treatments when compared to control group. Conclusion: In this rat model of intestinal obstruction, the treatment unith somatostatin analogue and omeprazole for 24 hours did not appear beneficial.

Çalışmanın amacı: Uzun etkili somatostatin analogu SMS 201-995 (Octreotide) ve bir proton pompa inhibitörü olan omeprazolun mekanik incebarsak obstruksiyonu oluşturulmuş rat modelindeki etkilerini incelemektir. Materyal ve metod: Deney için ortalama ağırlığı 150-200 gr olan erkek albino ratlar kullanıldı. Ratlar 24 saat süreyle aç bırakıldı. Eter inhalasyonu ile anestetize edildikten sonra laparotomi yapılarak terminal ileumda 10 cm.lik kapalı lup oluşturuldu. Ratlar 8 saatte bir subkutan olarak somatostatin analogu 50 µg/kg (n:10), salin 0.5 ml (n:10) ve omeprazol 7 mg/kg (tek doz verildi, sonraki dozlar salin olarak verildi) (n:10) almak üzere randomize edildiler. Tedavinin başlangıcından 24 saat sonra ratlar sakrifiye edildi ve barsakları çıkarıldı. Lup ağırlıkları ve luminal volümleri ölçüldü, pH ve elektrolit (sodyum, klor ve potasyum) analizleri yapıldı. Barsak lupları histolojik olarak incelendi. Bulgular: Kontrol grubu ile karşılaştırıldığında somatostatin analogu ve omeprazol tedavileri ile ince barsakta lup ağırlığı, luminal volüm, sodyum, klor, potasyum konsantrasyonları, pH değeri ve obstrukte barsakların makroskopik ve mikroskopik incelenmesinde anlamlı bir fark görülmedi. Sonuç olarak; İntestinal obstruksiyonlu bu rat modelinde 24 saatlik somatostatin analogu ve omeprazol tedavileri ile belirgin bir yarar sağlanmadı
