The tumor suppressor gene, pl6 is a cell cycle regulator that is frequently inactivated in ninny different types of malignancies. It was reported that pl6 may be a valuable biomarker for identification of elevated risk of cancer in smokers. The aim of present study was to investigate the possible role of pl6 as a biomarkcr by evaluating the expression of pi 6 in gingival epithelium of smokers and non-smokers, From smoker and non-smoker volunteers, 40 gingival biopsies were obtained from the mandibular molar attached gingiva. Gingival samples were stained with pl6 antibody by ininmnohistochcmistry. Hislopalhologieal evaluation was, done by light microscopy. The epithelial eelli that showed red nuclear/ cytoplasmic staining were accepted as positive. PI6 gene expression in smokers was higher (%60 in smokers, %4C> in nonsriiokers). The immunorcactivity of p 16 antibody was higher in males and older ages. Nevertheless, differences were not statistically significant.

Ornl kanserin de dahil olduğu birçok kanser gelişiminde, pl<j tümör supresör geninin inaklivasyonunun rol aldığı belirtilmektedir. Günümüzde p[6'njn özellikle sigara içen bireylerde, premaligıı veya malign ncoplazilerin erken iLinısiindn kullanılabilecek önemli bir biyomarker olabileceği düşünül m ekledir. Sigara, oral kanser gelişimindeki en büyük risk faktörlerinden biri olduğunu göre sigara içen bireylerin dişetinde pl6 inaktivasyonu saptanması, oral kavi-tede gelişebilecek olası bir malign veya prernalign lezyo-nun erken teşhirinde yararlı olabilir. Bu düşünceden yola çıkarak, sigara içen ve içmeyen toplam 40 adet gönüllü bireyden, mandibular posterior bölge yapışık dikelinden alınan örnekler immimhistokimyasai olarak plf> antikoru ile boyardı. İşık mikroskobunda değerlendirilen ve nükleer/ stoplazmik, kuvvetli kırmızı boyanan hücreler, pl6 proteini eksprese eden pozitif hücreler olarak kabul edildi. Toplam örneklerin %25'inde pl6 over ekspresyonu gözlendi. Sigara içen grupta pl6 ekspresyonu daha fazla bulundu (%60'ı sigara içenlerde, %40'ı sigara içmeyenlerde), Ayrıca ileri yaşlarda vc erkeklerde p!6 antikorunun ımmünrcaktivitesi daha yüksek saptandı. Ancak istatistiksel olarak gruplar arasında anlamlı bir farklılık izlenemedi,
